Abstract:  There is a possible relationship between migraine and hypercoagulability inducing factors, such as hyperhomocysteinemia. In this context, homocysteine (Hcy)-lowering vitamins (B6-folate-B12) may prove beneficial in the management-prophylaxis of migraine. We performed a systematic literature search in order to retrieve studies assessing the supplementation of B6, folate and B12 (alone or as adjunctive therapies) to migraine patients, as well as patients suffering from other primary headache disorders. MEDLINE, EMBASE, CENTRAL, Google Scholar, trial registries and OpenGrey were searched. Twelve relevant articles were retrieved. The management of acute migraine attacks with Hcy-lowering vitamins has not provided promising results (one randomized controlled trial-RCT-and one prospective uncontrolled trial). On the contrary, significant benefits were registered for the use of B6 alone, in combination with folate and in combination with folate and B12 in the prophylaxis of migraine with aura (MA) in adults compared to placebo (five RCTs, only one did not obtain significant results). Folate supplementation alone was not more efficacious than placebo (one RCT). Limited data for the prophylaxis of migraine without aura (MO) in children (two prospective uncontrolled trials) and adults (two prospective uncontrolled trials involving both MA and MO participants) impede the extraction of safe conclusions. An overall attractive safety profile was exhibited with gastrointestinal adverse events being the most common. Overall, a potential beneficial effect regarding the administration of B6, folate and/or B12 in the prophylaxis of MA in adults was indicated. Aim of this study was to evaluate functional and histopathological results of vitamins E and B12 on experimental rat nerve injury model.MATERIAL AND METHODS: This research included 32 Wistar Hannover rats. Sciatic nerves were crushed using an aneurysm clamp. Rats were divided into 4 groups, as group 0 (controls; no treatment), and groups B12, E, and B12+E. Rats were analyzed functionally, using sciatic functional index (SFI), and histopathologically.RESULTS: In functional analysis, it was determined that E was as influential as B12. Concomitant use of 2 vitamins was found to be more beneficial. SFI was significantly higher in B12+E group than B12 group, which indicated that E improved healing effects of B12. In histopathological evaluation, E was not effective in axonal degeneration (AxD) or edema/inflammation (EI) by itself. Although B12 was effective in treatment of EI, it was ineffective in treatment of AxD. However, combination decreased both AxD and EI, which showed; additive effects could reverse neurological injury when used together.CONCLUSION: B12 and E were effective in functional recovery of PNI. Neither vitamin B12 nor E was effective in treatment AxD; however, combination was effective in treatment of AxD. Results suggested that E was effective in treatment of PNI, especially when combined with B12. ABGE, vitamins D and C, and the Formulation suppressed LPS-induced gene expression of cyclooxygenase (COX)-2, tumor necrosis factor (TNF)-α, interleukin (IL)-6, nuclear factor-kB (NF-kB), and inducible nitric oxide synthase (iNOS) on mouse heart specimens. The beneficial effects induced by the extract could be related to the pattern of polyphenolic composition, with particular regard to gallic acid and catechin. The Formulation also increased fluorescence values compared to the vehicle, and it caused a significant membrane hyperpolarization of HASMCs compared to ABGE. To conclude, our present findings showed that ABGE, alone and in association with multivitamins, exhibited protective effects on mouse heart. Vitamins have important effects on cellular processes that are relevant for tumor development and progression.AIM: The present study explored the effect of pyridoxine or cobalamin supplementation on the viability and cell cycle progression of human glioblastoma cell line U-87 MG.METHOD: Cell cultures were treated with increasing concentrations of pyridoxine or cobalamin for 24-72 h. After supplementation, cell viability and cell cycle progression were assessed by spectrophotometry and flow cytometry. Analysis of Bcl-2 and active caspase 3 expression in supplemented cells was performed by western blot.RESULT: The results show that pyridoxine supplementation decreases cell viability in a dose and time dependent manner. Loss of viability in pyridoxin-supplemented cells is probably related to less cell cycle progression, higher active caspase 3 expression and apoptosis.
